Recommend Dilution	Western blotting: Use at 1/20-1/50 and anti-Rat-HRP. Immunocytochemistry: Use at 1/20-1/50 on Acetone Fixed Cytospin preparations.  Immunohistochemisty on Frozen Sections: 1/20 on Acetone Fixed Frozen sections can be followed by incubation with Rabbit anti-Rat Ig (1/25) and a monoclonal Rat APAAP complex (1/50). Alternatively, after incubation with M5I-10 (use 1/20) and washing, slides can be incubated with biotinylated Rabbit anti-Rat Ig (1/100) and streptavidin conjugated to horseradish peroxidase (1/500).

Specificity	This Monoclonal antibody M5I-10 reacts with an internal epitope of MRP5 (ABCC5), an approximately 160 kD transmembrane protein that is related to the multidrug resistance protein MRP1.  It also strongly reacts with the Human MRP5 protein.

Background	MRP5 (190-200 kDa) is closely related to MRP4, both lacking the first five membrane spanning regions. MRP5 is a GS-X multi specific organic anion pump (nucleotide analogs). MRP5 may transport DNP-GS and may be inhibited by certain inhibitors of organic anion transport (sulfinpyrazone). MRP5 may also transport organic anions with the anionic moiety of phosphate/phosphonate group, a function which provides the ability to resist against anti cancer drugs 6-MP and thioguanine as well as the anti-HIV drug PMEA.
