Applications Protocol: Activity Assay
1. Prepare a 1 ml reaction cocktail into a suitable container: The final concentrations are 5mM Tris, 63mM sodium phosphate, 1mM DTT, 0.1% (w/v) insulin, 2mM EDTA.
2. Add 4ug of recombinant thioredoxin into reaction cocktail and mix immediately.
3. Incubate at 25°C for 20 minutes.
4. Record the increase in A650nm for 15 minutes
Protein Description Recombinant human thioredoxin-1 was overexpressed in E. coli and purified by using conventional chromatography techniques.
Storage Store undiluted at 2-8°C for up to two weeks or (in aliquots) at -20°C or -70°C for longer.
Avoid repeated freezing and thawing.
Stability Shelf life: one year from despatch.
Bioactivity Specific activity is >150 A650/cm/min/mg, obtained by measuring the increase of insulin precipitation in absorbance at 650 nm resulting from the reduction of insulin. Please refer to our activity assay protocol below.

The encoded protein is active in the reversible S-nitrosylation of cysteines in certain proteins, which is part of the response to intracellular nitric oxide. This protein is found in the cytoplasm.
